Cytokine secreted predominantly by activated T-lymphocytes as well as mast cells and osteoblastic cells that controls the production and differentiation of hematopoietic progenitor cells into lineage-restricted cells (PubMed:2556442). Stimulates also mature basophils, eosinophils, and monocytes to become functionally activated (PubMed:10779277, PubMed:32889153). In addition, plays an important role in neural cell proliferation and survival (PubMed:23226269). Participates as well in bone homeostasis and inhibits osteoclast differentiation by preventing NF-kappa-B nuclear translocation and activation (PubMed:12816992). Mechanistically, exerts its biological effects through a receptor composed of IL3RA subunit and a signal transducing subunit IL3RB (PubMed:29374162). Receptor stimulation results in the rapid activation of JAK2 kinase activity leading to STAT5-mediated transcriptional program (By similarity). Alternatively, contributes to cell survival under oxidative stress in non-hematopoietic systems by activating pathways mediated by PI3K/AKT and ERK (PubMed:27862234).

Phenotypic changes associated with continuous long term in vitro expansion of bone marrow-derived mesenchymal stem cells

In vitro expansion of mesenchymal stem cells is necessary to obtain a higher cell number for clinical applications. However, long-term expansion can produce significant phenotypic changes on these cells, decreasing their therapeutic utility. Therefore, understanding the phenotypic changes that long-term expansion triggers in mesenchymal stem cells will allow for better and more consistent cell therapy results. Here, we evaluate the phenotypic changes caused by continuous passaging through colony forming unit-fibroblast assay, senescence beta-galactosidase staining, morphology examination, secretome analysis, surface marker expression, protein quantification, osteogenic and adipogenic differentiation, and CD4⁺ T lymphocyte immunosuppressive potential. Long-term in vitro culture decreases mesenchymal stem cell osteogenic potential and self-renewal, increases cell size, and senescence, but does not consistently affect adipogenic differentiation. Surface marker expression remains similar for positive and negative markers, while secretory phenotype shifts with decreased p14ARF, MMP-3, p21 Waf1/Cip1,ENA-78, GCP-2, GROα, IL-3, IL-7, IL-8, RANTES, TNFβ, and VEGF-A expression, and increased p53, p16 INK4a, MCP-1, and SDF-1 expression. Immunomodulatory potential remains unchanged. These findings can help better understand the phenotypic changes that mesenchymal stem cells undergo while expanded in vitro.

Refining the interactions between microglia and astrocytes in Alzheimer’s disease pathology

Microglia and astrocytes are central to the pathogenesis and progression of Alzheimer's Disease (AD), working both independently and collaboratively to regulate key pathological processes such as β-amyloid protein (Aβ) deposition, tau aggregation, neuroinflammation, and synapse loss. These glial cells interact through complex molecular pathways, including IL-3/IL-3Ra and C3/C3aR, which influence disease progression and cognitive decline. Emerging research suggests that modulating these pathways could offer therapeutic benefits. For instance, recombinant IL-3 administration in mice reduced Aβ plaques and improved cognitive functions, while C3aR inhibition alleviated Aβ and tau pathologies, restored synaptic function, and corrected immune dysregulation. However, the effects of these interactions are context-dependent. Acute C3/C3aR activation enhances microglial Aβ clearance, whereas chronic activation impairs it, highlighting the dual roles of glial signaling in AD. Furthermore, C3/C3aR signaling not only impacts Aβ clearance but also modulates tau pathology and synaptic integrity. Given AD's multifactorial nature, understanding the specific pathological environment is crucial when investigating glial cell contributions. The interplay between microglia and astrocytes can be both neuroprotective and neurotoxic, depending on the disease stage and brain region. This complexity underscores the need for targeted therapies that modulate glial cell activity in a context-specific manner. By elucidating the molecular mechanisms underlying microglia-astrocyte interactions, this research advances our understanding of AD and paves the way for novel therapeutic strategies aimed at mitigating neurodegeneration and cognitive decline in AD and related disorders.

Enhanced Siglec-8 and HLA-DR and reduced CRTH2 surface expression highlight a distinct phenotypic signature of circulating eosinophils in atopic dermatitis

AbstractAtopic dermatitis and other type 2 immune response diseases are often linked to elevated eosinophil levels in the blood. Although the role of eosinophils in atopic dermatitis pathophysiology is suspected, it remains unclear. The development of new treatments targeting the type 2 response, particularly cytokines involved in eosinophil activation and chemotaxis, makes it necessary to identify potential eosinophil profiles in atopic dermatitis that may respond to these treatments. A prospective study was conducted comparing blood eosinophil phenotypes in patients with moderate to severe atopic dermatitis (n = 19) without recent systemic treatment to healthy individuals (n = 19). The primary outcome was the membranous phenotypic signature of eosinophils, assessed by flow cytometry. Most patients with atopic dermatitis (84%) had early onset in childhood, a severe disease (mean SCORing Atopic Dermatitis of 57.5), and elevated blood eosinophil counts (310 per µL in atopic dermatitis vs 120 in healthy individuals, P < 0.0001). Patients with atopic dermatitis exhibited lower CRTH2 on eosinophils but higher levels of human leukocyte antigen–DR isotype and Siglec-8 compared to healthy individuals. Other surface proteins showed no significant differences. Clustering analysis confirmed increased Siglec-8 in patients with atopic dermatitis. Additionally, patients with atopic dermatitis had higher serum levels of type 2 immune response markers such as eotaxin-2, IL-5, IL-3, and TARC. Circulating eosinophils in patients with atopic dermatitis show a distinct phenotypic profile, suggesting a role in atopic dermatitis pathophysiology and potential involvement in differential treatment responses.

Versant startup Granite debuts with $100M and two immune drugs

Venture capitalists have been scouring the globe in search of better immunology drugs. Biotech startup creator Versant Ventures is among them, having formed and funded several companies in recent years looking to change the way certain inflammatory diseases are treated.Versant isnt done with that effort. On Thursday, the firm revealed its latest foray into immunology research a startup called Granite Bio thats already secured $100 million in funding and has two drugs either in, or nearing, clinical testing.Granite is a product of Ridgeline Therapeutics, part of an initiative Versant hatched more than a decade ago to help turn scientific discoveries into new companies. Operating in stealth mode, the company raised a $30 million Series A led by Versant and Novartis venture arm in 2021 and a $70 million Series B from Forbion and Sanofis venture fund two years later.The startup is working on two antibody drugs for inflammatory conditions. One, dubbed GRT-001, is being developed for inflammatory bowel disease and is currently in Phase 1 testing in healthy volunteers, with a trial in ulcerative colitis patients expected later this year. Another, GRT-002, is seen as a new way to potentially treat itch and allergies and should start its first trial in 2026.GRT-001 homes in on white blood cells called pro-inflammatory monocytes, which are part of the immune systems response to infections and inflammation. According to Granite CEO Patrick Loustau, this approach targets the “cellular source“ of IBD and has the potential to “reset“ patients' immune systems, which could lead to better and longer-lasting results than existing therapies.Monocyte depleters could have a similar impact on the innate immune system as B-cell targeting drugs appear to have on the adaptive part of the bodys defenses, giving them the potential to treat a range of inflammatory conditions,Loustau wrote in an email to BioPharma Dive.Granites other drug, GRT-002, homes on IL-3, a different inflammatory cytokine than the ones targeted by drugs like Dupixent and Cosentyx. Granite, in a statement announcing its launch, noted how IL-3 is a key player in autoimmunity as well as a kind of immune response called type 2 inflammation.Versant has started up several immune disease-focused biotechsof late. In 2024 alone, the firm formed Santa Ana Therapeutics, a developer of targeted immune therapies, as well as Borealis Biosciences, which is focused on medicines for inflammatory kidney diseases.Immunology investments more than doubled last year, surging to about $3.1 billion among the private rounds involving the prominent venture firms tracked by BioPharma Dive. Theyre on a similar trajectory in 2025, with about $1 billion raised so far. '

癌症中的细胞因子

细胞因子是由免疫细胞（如单核、巨噬细胞、T细胞、B细胞、NK细胞等）和某些非免疫细胞（内皮细胞、表皮细胞、纤维母细胞等）经刺激而合成、分泌的一类具有广泛生物学活性的小分子蛋白质。具有调节固有免疫和适应性免疫、血细胞生成、细胞生长、APSC多能细胞以及损伤组织修复等多种功能。细胞因子可被分为白细胞介素(IL)、干扰素(IFN)、肿瘤坏死因子(TNF)、集落刺激因子(CSF)、趋化因子、生长因子(GF)等。 01 细胞因子发挥作用的方式 02 细胞因子的作用特点多效性：一种细胞因子可以对不同细胞发挥作用；重叠性：两种或两种以上的细胞因子具有同样或类似的生物学作用；协同性：一种细胞因子可增强另一种细胞因子的功能；拮抗性：一种细胞因子可抑制另一种细胞因子的功能；多相性网络性效应 03 细胞因子是信号因子增强免疫细胞效应为了维持机体的生理平衡，抵抗病原微生物的侵袭，防止肿瘤发生，机体的许多细胞，特别是免疫细胞合成和分泌许多种细胞因子，并且通过细胞因子彼此沟通。细胞因子是细胞释放到血液中的小分子蛋白质，可以令免疫细胞冲到感染部位、吞噬遭到损伤的细胞，甚至穿透血管壁，细胞因子还可以引发炎症，令被破坏的机体肿胀、发热以及疼痛。细胞因子在细胞之间传递信息，调节细胞的生理过程，提高机体的免疫力，在免疫过程中，免疫细胞可以通过分泌细胞因子，招募更多的免疫细胞，更快地清除病原体。因此，我们认为细胞因子是一种“信号分子“！ 04 细胞因子的应用领域 1、治疗疾病目前，利用基因工程技术产生重组的细胞因子作为生物应答调节剂，已用于临床治疗肿瘤、感染、造血功能障碍等，并获得了较好的疗效。细胞因子已成为新一代的治疗药物，其低剂量即可发挥药效作用，且不良反应较轻，是一种全新的生物制剂，也是对一些疑难杂症不可缺少的治疗手段。目前已获批准的药物有IFN-α、IFN-β、IFN-γ、IFN-ε、PO、GM- CSF、G- CSF、IL- 3；临床试验的药物有IL- 1、IL- 3、IL- 4、IL- 6、IL- 11、M- CSF、SCF、TGF-β等。其通过各种途径而使体内细胞因子水平增加，达到治疗疾病的目的。临床治疗效果较好的有IFN、IL- 2、TNF、CSF。细胞因子阻断和拮抗疗法是抑制细胞因子的产生和阻断细胞因子与其受体结合，以及受体后信号的传导过程，使细胞因子的病理性作用难以发挥。 2、检测对细胞因子的检测不仅是基础免疫研究的有效手段，同时在临床对疾病诊断、疗效判断及细胞因子治疗检测方面具有重要价值。但是，由于细胞因子在体内的含量甚微，对细胞因子检测的实施较为困难，而且采用不同方法所得的结果差异也较大，其准确性难以判断。目前检测方法有：⑴生物学检测法。包括细胞增殖法、靶细胞杀伤法、细胞因子诱导的产品分析法和细胞病变抑制法。⑵免疫学检测法。⑶分子生物学方法。以上检测方法，各有其优缺点。生物学检测法为常用方法，比较敏感、可靠，但检测时间较长，步骤繁多，不易掌握；免疫学检测法较简单、迅速，且敏感度低于生物学检测；分子生物学检测法是检测基因的表达，主要用于对机制的探讨。 3、美容医学皮肤生长期一般在25岁左右就结束了，也就是说25岁是年轻肌肤与老化肌肤的分水岭，之后生长与老化是同时进行的。人体内细胞因子的自然生理含量极少，由于其生理功能和生物学效应贯穿于人体生长发育的全过程，故其扮演着重要角色。当前，细胞因子是医疗美容市场应用较广泛的物质之一。目前应用生物美容的生长细胞因子有EGF、FGF、KGF，以及其他细胞因子(VEGF、HGF、IGF- 1)。 ⑴表皮生长因子(EGF)。于1962年由Montalcini和Cohen发现，并于1986年获得了诺贝尔奖，从此揭开了皮肤衰老之谜。因其能促使皮肤的年轻化，故被称为“美丽因子”。主要作用是调节表皮与上皮细胞生长及分化增殖，促进毛细血管生长，改善细胞微环境，从而保持细胞的活性等。 ⑵成纤维细胞生长因子(FGF)。包括酸性成纤维细胞生长因子(aFGF)和碱性成纤维细胞生长因子(bFGF)。这两种细胞因子均能促进成纤维细胞生长，提高皮肤合成胶原蛋白的功能，从本质上改善了皮肤的状况。其主要作用是修复损伤、去皱、美白、祛斑、防粉刺等。 ⑶角质细胞生长因子(KGF)。该生长因子属于FGF家族。由间充质干细胞分泌，并通过旁分泌途径来刺激上皮细胞增殖，促进且维持人角化细胞、表皮细胞及上皮细胞的增殖、迁移分化和存活等，并能促使皮肤毛囊、皮脂腺前体细胞增强和分化，激发头皮毛囊的发育，预示生发的潜在应用。 ⑷其他。血管内皮细胞生长因子(VEGF)能促进血管内皮细胞增殖及血管生成，增加血管通透性，对改善皮肤微环境循环并增强皮肤细胞的合成代谢起着重要作用；干细胞生长因子(HGF)是由人脑垂体前叶分泌的。自1985年罗德曼将HGF应用于临床30余年来的研究表明，IGF- 1、脑垂体前叶分泌1的HGH能独立存在2～4 min，并由肝脏迅速转化为胰岛素样生长因子1(IGF-1)在体内发挥作用。IGF- 1与血小板衍生生长因子(PDGF)有协同作用，能增加表皮和内皮的再生。 05 52中细胞因子的来源和功能临床可以从细胞因子水平，大致了解哪些免疫细胞参与了反应，产生了什么后果，从而与临床表现进行联系；科研可以从中了解作用机制和参与的细胞组成。 Ref. 来源：细胞王国推文用于传递知识，如因版权等有疑问，请于本文刊发30日内联系医药速览。原创内容未经授权，禁止转载至其他平台。有问题可发邮件至yong_wang@pku.edu.cn获取更多信息。 ©2021 医药速览保留所有权利往期链接 “小小疫苗”养成记 | 医药公司管线盘点人人学懂免疫学 | 人人学懂免疫学（语音版）综述文章解读 | 文献略读 | 医学科普 | 医药前沿笔记 PROTAC技术 | 抗体药物 | 抗体药物偶联-ADC 核酸疫苗 | CAR技术 | 化学生物学温馨提示医药速览公众号目前已经有近12个交流群（好学，有趣且奔波于医药圈人才聚集于此）。进群加作者微信（yiyaoxueshu666）或者扫描公众号二维码添加作者，备注“姓名/昵称-企业/高校-具体研究领域/专业”，此群仅为科研交流群，非诚勿扰。简单操作即可星标⭐️医药速览，第一时间收到我们的推送 ①点击标题下方“医药速览” ②至右上角“...” ③点击“设为星标
